摘    要:以水稻粳稻品种空育131、垦稻12为材料,用农杆菌共转化的方法将质粒p1300和p13W8导入受体材料。结果显示:两种混合共转化菌液按体积比1∶1时共转化效率最高,植株转化效率达4.08%和2.63%。将转化植株进行PCR鉴定,13株转基因植株均能检测到质粒p1300的信号,4株检测到质粒p13W8的信号,反义蜡质基因的阳性株占转基因植株总数的30.77%。

Abstract:Both of the plasmid p1300 and plasmid p13W8 were introduced into Japonica Rice Kongyu 131 by Agrobacterium-mediated co-transformation,the same two plasmid were introduced into Kendao 12 by the same way. The results showed that the co-transformation efficiency was highest when two kinds of Agrobacterium was 1 : 1 volume,and the co-transformation efficiency were 4.08% and 2. 63%, respectively. The transformed plants were screened by PCR method,specific fragments of plasmid p1300 could be screened in 13 transgenic lines,specific fragments of plasmid p13W8 could be screened in 4 transgenic lines. Positive Antisense-Waxy Gene plants were 30.77% of all transformed plants.
