梨试管苗的生根

摘    要:采用三种不同生根法,成功地诱导梨砧木BP10030试管苗生根.试管生根法最适的吲哚丁酸(IBA)浓度为2mg·1~(-1),生根率、平均根数和根长度分别为95%、7.4和3.4cm.用2—8mg·l~(-1)的低浓度IBA浸枝处理,生根率均在90%以上,其中以4mg·l~(-1)的IBA促根效果最佳.用10—500mg·l~(-1)的高浓度IBA浸枝处理,生根较差.尽管试管生根法容易诱导试管苗形成不定根,但是不定根形成部位产生大量愈伤组织,试管苗移栽成活率低,初期生长缓慢.采用低浓度IBA浸枝生根法,不定根形成部位不产生愈伤组织,移栽成活率高,试管苗初期生长快.

Abstract:Rooting of microcuttings of the pear rootstock BP10030 were successfully achieved by using three different rooting methods. When rooting in vitro, the optimum IBA concentration was found to be 2mg · l-1 under which condition 95% of the microcuttings produced roots, with mean number of roots of 7. 4 by each and with mean root length of 3. 4 cm. More than 90% of microcuttings produced roots when soaking treatment was supplied using 2-8mg · l-1 IBA with the highest rooting obtained at 4mg · l-1 IBA. Lower rooting was found in microcuttings treated by 10-500mg · l-1 IBA. Although adventious roots may be easily induce in vitro culture, callus usually formed at the base of microcuttings leading to low survival when transplanting and poor growth of plantlets. No callus formed when microcuttings were rooted by IBA soaking method and plantlets acclimated easily for transfer to greenhouse conditions and grew faster initially.
