非洲紫罗兰体细胞胚胎发生过程中生化变化的研究

摘    要:鉴定了非洲紫罗兰叶片培养体细胞胚胎发生的发育进程,测定了各发育时期核酸、蛋白质和淀粉的含量.叶片接种后1~12d为脱分化期,13~16d为胚性愈伤组织形成期,17~31d为原胚形成期,32~48d为胚状体发育期.脱分化过程中的DNA,RNA和蛋白质含量低于再分化过程,淀粉含量高于再分化过程.胚性愈伤组织形成过程中,RNA和酸溶性蛋白质含量上升,淀粉含量下降.原胚形成和胚状体发育过程中,DNA含量上升,淀粉含量进一步下降.

Abstract:The developmental stages of somatic embryogenesis were determined and the contents of nucleic acid,protein and starch at each stage were measured during leaf cultureof Saintpaulta ionantha.It was found that the dedifferentiation period lasted from1 to 12 days after inoculation,the em bryonic callus formation period Was from 13 to 16 days,the pro embryo formation period was between 17 and 31 days and the embryonic development period was from 32 to 48days.The contents of DNA, RNA, and Protein at dedifferentiation Period were lower thanthat at redifferentiation period, while the content of starch at dedifferentiation period washigher than that at redifferentiation period.During embryonic callus formation,the contentsof RNA and acid dissolved protein inereased,and those of starch decreased.During embryonie development,the content of DNA increased,and that of starch deereased further.
