摘    要:试验结果表明,处于对数生长期、再生能力强的培养细胞有利于原生质体分离和培养。原生质体分离和初期培养的适宜渗透浓度为0.5M;固体包埋法的效果明显优于液体培养,前者分裂率为后者的3.8倍,达13.8%;外源激素浓度以 B_5基本培养基添加 NAA 2.0mg/L,BA 2.0mg/L,原生质体分裂率最高;葡萄糖取代常规稳压剂甘露醇,效果显著,细胞分裂率是后者的2.6倍。

Abstract:Effects on grape protoplast isolation and division of several factors such as source of ma-terial,concentration of auxins,ulturing methods,different kinds and concentrations ofosmotica were discussed in this paper.The result showed that cells in their logarithm growthperiod with high potency of regeneration were suitable for protoplast isolatiaon and culture.The optimum osmoticum was 0.5M for protoplast isolation and in the early period of culture.The percentage of protoplast division in gum-solidified medium was 13.8%,which was 3.8times as high as that in liquid medium.B_5medium supplemented with NAA 2.0mg/L,BA2.0mg/L hit the highest percentage of protoplast division.Using glucose as an osmoticuminstead of the commonly used manni(?)ol showed much higher percentage of protoplast divi-sion,which was 2.6 times as high as that of mannitol.
