摘    要:以蓝果忍冬品种邱雷姆和娜雷姆无菌苗叶片为外植体,添加一种或多种生长调节剂诱导愈伤组织,结果表明,不同生长调节剂对蓝果忍冬叶片愈伤组织诱导影响不同,单独添加2,4-D(0.5~2 mg·L-1)可有效诱导蓝果忍冬叶片产生愈伤组织,出愈率达100%;单独添加6-BA和TDZ无法诱导蓝果忍冬叶片产生愈伤组织;6-BA(1~2 mg·L-1)与2,4-D(0.2、0.5、1.0 mg·L-1)配合使用可改善愈伤组织状态,增加绿色致密型愈伤组织,愈伤量增多;添加6-BA、IBA、KT组合的三种生长调节剂,品种邱蕾姆出愈率和愈伤量优于娜蕾姆,三种添加方式比较而言,6-BA与2,4-D组合诱导效果相对较好。研究可为蓝果忍冬组培快繁体系建立奠定基础。

Abstract:In this study, leaves of sterile seedlings of blue honeysuckle varieties Qiuleimu and Naleimu were used as explants to induce callus by adding one or more growth regulators. The results showed that different growth regulators had different effects on the callus induction of blue honeysuckle. The addition of 2,4-D alone (0.5-2 mg·L-1) could effectively induce the callus from blue honeysuckle leaves, and the callus ratio reached 100%; addition of 6-BA and TDZ could not induce the callus from blue honeysuckle leaves;The use of 6-BA (1-2 mg·L-1) in combination with 2,4-D (0.2, 0.5, 1.0 mg·L-1) could improve callus status and increase the rate of green dense callus, and the amount of callus had also increased; with the addition of three growth regulators of 6-BA, IBA and KT, Qiuleimu induction rate and amount of callus were better than those of Naleim. Comparing the three methods of addition, the combination of 6-BA and 2, 4-D had a relatively good induction effect. The study laid a foundation for the establishment of the tissue culture and rapid propagation system of blue honeysuckle.
