大豆TST基因的生物信息学及表达模式分析

摘    要:TST为典型膜结合蛋白,具有高度保守序列,广泛存在于植物组织和细胞中。文章根据NCBI网站、Phytozomes数据库和MEGA软件分析大豆基因组中TST同源基因生物信息,发现大豆相关数据库中有8个大豆TST相关基因拷贝,其中5个TST基因与拟南芥和甜菜亲缘关系较近;通过实时荧光定量PCR分析大豆TST基因时空表达模式,确定GmTST2.1基因发挥拷贝作用;克隆GmTST2.1基因并分析其生物信息发现,GmTST2.1蛋白质由734个氨基酸组成,PI为5.1不稳定疏水性蛋白质,二级结构主要由α-螺旋、无规则卷曲和片层结构组成,有典型N端跨膜结构域-中间胞质区-C端跨膜区结构域特征。研究分析GmTST基因生物信息学和表达模式,为进一步验证GmTST基因生物学功能提供借鉴和理论基础。

Abstract:Tonoplast sugar transporter (TST) is a typical membrane-binding protein with highly conserved sequence,which widely distributes in the tissues and cells of plants.In the present study,bioinformatical analysis of TST homologous throughout soybean genome was analyzed through NCBI website,Phytozomes databank and MEGA softwares,a total of eight soybean GmTST genes in soybean genome were identified.According to TST sequence and phylogenetic relations,five GmTST genes were closely related to those of Arabidopsis thaliana and Beta vulgaris.The expression patterns of five GmTST genes in different soybean tissues were analyzed by real-time quantitative PCR.The results showed that GmTST2.1 gene played a major role.So,GmTST2.1 gene was cloned and then bioinformatically analyzed.GmTST2.1 was a 734 amino acids hydrephobic protein (PI 5.1).The main structure elements of the deduced protein included the typical N-terminal transmembrane domain-Intermediate cytoplasmic region and C-terminal transmembrane domain,α-helix,β-sheet and random coil.The result of this study would provide reference and theoretical basis for further validation of GmTSTgene function.
