杂交水稻中耐热基因的筛选和验证

摘    要:分别以Ⅱ优838及其两个亲本的处理组和对照组开花当日的剑叶为材料,应用抑制消减杂交方法筛选得到一批耐热相关基因,选取其中6种和耐热密切相关的基因,在亲本和子代中分别检测其表达情况,结果显示:6种基因在高温处理组中的表达量都明显上调,且在子代中的表达高于亲本.这不仅证实了获得的差异表达基因的可靠性,还说明这些基因能稳定的遗传给后代.

Abstract:Sword leaves of heat treatment group and control group of ⅡYOU838 and its homozygote parents were collected on their blossoming day to screen the differentially expressed sequences related to the heat resistance by suppression subtractive hybridization. Several differentially expressed sequences were selected,of which functions might cover a variety of aspects. To confirm the differential expressions of the genes,6 index genes for plant heat resistance were chosen for RT-PCR and northern hybridization analysis in parents and their filial generations. The results showed that heat treatment could up-regulate the expression of all 6 genes,and this furtherly validated the differentially expressed sequences screened from SSH library. The expression of these genes also increased in filial generations after heat treatment,indicating that these genes could be inherited and play important roles for rice under high temperature stress in filial generations.
