野生茄转化酶抑制子基因 StINH1的克隆与序列分析

摘    要:以1个受黄萎病菌诱导的“托鲁巴姆”下调EST为种子序列,在NCBI进行同源搜索,经人工拼接、RT PCR克隆与序列分析验证,获得野生茄“托鲁巴姆”转化酶抑制子基因(INH)的全长cDNA序列,命名为 StINH1。该基因的开放阅读框全长531 bp,编码176个氨基酸,与电子克隆获得的序列完全相同。编码蛋白的分子质量为19.916 ku,理论等电点为5.14。序列分析表明,该基因属于植物PMEI超家族,编码的前体蛋白含有1个拥有19个氨基酸的前导肽,同时含有多个磷酸化位点。表达模式分析表明,在黄萎病菌侵染后, StINH1在“托鲁巴姆”根中呈下调表达。

Abstract:In this study, a down regulated EST in Solanum torvum induced by Verticillium wilt was used as a querying probe to blast the EST database in GenBank. Based on the assembled homologous cDNA sequences, RT PCR and sequence analysis, a full length cDNA sequences of invertase inhibitor gene (INH) was confirmed, designated StINH1. It contains a 531 bp open reading frame (ORF) that encodes a 176 amino acid protein. The coding protein has a molecular mass of 19.916 ku and a theoretical isoelectric point of 5.14. Sequence analysis showed that StINH1 belongs to plant PMEI superfamily, and codes a precursor protein containing a 19aa leader peptide and multiple phosphorylation sites. The expression pattern indicated that StINH1 was down regulated while Solanum torvum was infected by Verticillium wilt.
