胶束增敏荧光法测金银花中黄酮的含量

摘    要:选用微波提取法提取出了金银花中总黄酮,通过化学方法对金银花中的总黄酮进行了定性研究。最后用荧光光度法对金银花中的总黄酮进行了含量测定,研究了稳定性实验,干扰试验,以及表面活性剂、温度、pH等对含量测定的影响,选出最佳测定条件为静置15 min,室温情况下测定较准确,缓冲溶液的pH为11,为了增强荧光强度加入的表面活性剂为环糊精,然后在最佳测定条件下研究了回收率实验,测得回收率为52.93%。最后测定了金银花中的黄酮的含量为6.10%。实验结果表明,该方法灵敏度高,线性范围宽,数据重现性好,操作简便,可广泛用于植物中黄酮测定,具有很好的应用前景。

Abstract:Total flavonoids from honeysuckle was extracted by microwave methods.the qualitative analysis of flavonoids from honeysuckle was achieved by chemical reaction.the total content of flavonoids from honeysuckle was investigated.And the optima condition as the stability,interference testing,surface active agent,temperature and pH of the method was studied.the results showed that the best condition was static for 15 minutes,at room temperature,the buffer solution'pH equal to 11,adding surfactants CD in order to enhance the fluorescence intensity,and the recovery test was measured reach to52.93%.And the content of total flavonoids from honeysuckle was 6.10%.Experimental results showed the method has high sensitivity,wide linear range,the data reproducible,easy to operate,can be widely used to determinate the content of flavonoids.
