阔叶十大功劳叶柄愈伤组织诱导及试管苗培养的研究

摘    要:以大连市引种的阔叶十大功劳嫩叶柄为外植体,采用组织培养方法研究其快繁技术,结果表明:愈伤组织诱导的初代和继代理想培养基是MS+KT 0.6 mg·L-1+2,4-D 2.5 mg·L-1;不定芽分化的理想培养基是1/4MS+ZT0.4 mg·L-1+NAA 0.1 mg·L-1;不定芽生根的理想培养基是1/3MS+IAA0.5mg·L-1;试管苗生根继代培养形成的试管苗生长旺盛,生根率为94.6%,生根数为7.2条,每代的繁殖系数为2.8。试管苗移栽平均成活率为89.8%,生长旺盛,在大连地区可正常越冬,翌年即可开花。

Abstract:By using tissue culture technique,the tender petiole of Mahonia bealei was used as explant.Researches on callus induction and differentiation,rooting of adventitious buds,rooting and transplanting of tube seedlings,and clone establishment of Mahonia bealei were conducted in this paper.The results showed that MS+KT 0.6 mg·L-1+2,4-D 2.5 mg·L-1 was the optimum medium for callus induction and subculture.The optimum medium for callus differentiation was 1/4MS+ZT0.4 mg·L-1+NAA 0.1mg·L-1.After dealt with NAA(concentration of 20 mg·L-1) for 3 minutes,the adventitious buds could be easily induced to rooting in the medium of 1/3MS+IAA0.5 mg·L-1.The rooting rate was 94.6% with 7.2 roots on average.The survival rate of transplanted tube seedlings was 89.8%.Tube seedlings could overwinter in Dalian,growing vigorously and flowering in the following year.
