多蕊金丝桃的组织培养

采用西藏林芝地区野生花卉多蕊金丝桃的茎断为外植体,试验结果表明①对比野外直接采用外植体和经室内黄化处理的外植体组织培养效果分析发现,经黄化处理的外植体污染率低,但分化明显没有野外直接采用的外植体强.②在MS培养基上,当NAA浓度为0.5mg/kg时,BA浓度取1.0～4.0mg/kg时,多蕊金丝桃的芽诱导率较高,而且去除顶端是促进芽发生的有效途径.③采用MS+IBA3.0mg/kg作为壮苗培养时最为有效,产生的芽健壮.在再一次的培养中观察到,在MS+IBA3.0mg/kg上培养的芽丛再次转接到MS+NAA0.5mg/kg+BA1.0mg/kg上后才能继续扩繁.④在1/2MS+IBA2.0mg/kg+BA0.5mg/kg+KT6.0mg/kg+0.2%AC(活性碳)培养基上生根效果明显.出瓶成活率达95%以上.

THE TISSUE CULTURE OF HYPERICUM HOOKERIANUM

Its rapid propagation of was studied by adjusting the kinds and concentrations of hormones in the basic MS, 1/2MS, MS\|Anderson medium. Acccording to the exprement, the efficient medium for the first and multiplated culture is MS. Compared with the shoots yellowed in the room, the shoots dug from wildness produce more shoots, but the polluted rate raises.In the first culture, when the concentrations of NAA is 0 5 mg/kg, and that of BA is 1.0 to 4.0 mg/kg, An average of 4.3 to 7.7 mg/kg flod multiplication rate of shoots is produced after 2 month of culture. And the shoots cut tips could make it an average of 35 to 11.7 mg/kg flod. In the strong sprout culture, the prefect results is obtained with IBA at 3.0 mg/kg. For multiplated more shoots, the shoots which had been stronged should be cultured in first culture medium again. The havested shoots are rooted in 1/2MS IBA at 2.0 mg/kg BA at 0.5 mg/kg KT at 6.0 mg/kg 0.2% activated charcoal. The rooting rate is about 93.75% after 1 monthes of culture, the medium for plantlets is pine sawdust which had killed mushroom by hight temperature.The survival rate is 95%.
