摘    要:选用棉花黄萎病菌代表性的菌系Va(Veticlliumalboatrum的一个菌株)和Veticilliumdahliae的落中型菌系VD-8,T9及非落 叶型菌系Jy,以Ke作为实验对象,研究了CTAB和氯化苄2种提取方法的优劣。

Abstract:The study on the methods of DNA extraction showed that CTAB method is better than PhCH 2Cl method. DNA extracted by CTAB method has longer fragment, higher purity, better stability, and less pollution. So it is suitable for the conventional DNA extraction of cotton Verticillium wilt pathogen. The primers screening of RAPD analysis has been performed with 40 primers (OPY, OPB) and 16 primers amplified polymorphic DNA patterrn. RAPD analysis of different single spores also proves that the genetic variation may occur among different single spore from one species.
