荻幼穗愈伤组织诱导及无性系的建立

摘    要:以获幼穗为外植体诱导出愈伤组织,并建立了体细胞无性系,获得了大量试管苗。幼穗愈伤组织诱导率与发育时期及转管培养降低紫色分泌物浓度有密切关系。BA和2,4-D配合使用有利于胚性愈伤组织的形成,在2,4-D 2mg/L+BA(或KT)0.1mg/L的培养基上。从愈伤组织表面产生浅黄色、质地致密的胚性愈伤组织。愈伤组织转入无激素培养基上培养,也有利于胚性愈伤组织的形成。为了形成大量试管苗,进行了愈伤组织的继代培养和试管苗的增殖研究,再生能力通过无性系得到保持。

Abstract:Calli have been induced from the young ear explants of Amur sil-vergrass. A somaclone has been established and a lot of test-tube shoots havebeen obtained. The induced percentage of calli is closely correlated to the deve-lopmental period and reduction of purple secretion by transfer-tube culture and2, 4-D are conductive to the formation of calli. The embryonal calli formed onthe callus surface cultured on MS medium with 2, 4-D 2 mg/L BA (or KT) 0.1mg/L are light yellow in colour, fine and close in texture. Transfer of calliinto MS medium without hormone is beneficial to the formation of embryonalcalli. Callus subculture and the shoot proliferation have been carried out to ob-tain a quantity of tube shoots. The regenerative ability has been retained by es-tablishing a somaclone.
