泰山松花粉多糖和乳酸杆菌对禽波氏杆菌OMP亚单位疫苗免疫增强的协同作用

摘    要:采用改进的水提醇沉法提取泰山松花粉粗多糖,并制备禽波氏杆菌OMP亚单位疫苗。取1日龄SPF雏鸡300只,随机分为6组,I~V组分别免疫含有松花粉多糖100g/I。的禽波氏杆菌OMP亚单位疫苗,Ⅵ组只免疫禽波氏杆菌OMP亚单位疫苗,I、Ⅱ组在前2周分别口服高、低剂量的乳酸杆菌活菌菌液1mL,HI、IV分别口服高、低剂量的乳酸杆菌热灭活菌菌液1mL。在首免后,每周采取血液和小肠,用间接ELISA法检测血清抗体水平、MTT法检测外周血淋巴细胞转化率、全自动血细胞分析仪测定外周血淋巴细胞比率、ELISA法检测小肠sIgA含量和IL-2水平。结果表明,I~Ⅳ组抗体水平、IL一2含量、淋巴细胞比率、淋巴细胞转化率、sIgA含量显著高于V组、Ⅵ组(P%0.05);V组各检测指标显著高于Ⅵ组(P〈O.05);乳酸杆菌活菌组各检测指标比灭活菌组略高,高剂量组略高于低剂量组。因此,泰山松花粉多糖作为免疫佐剂能显著提高疫苗的免疫效果,同时1:3服乳酸杆菌能进一步增强动物对疫苗的免疫应答能力,口服乳酸杆菌活菌与口服热灭活菌效果差异不显著,泰山松花粉多糖和乳酸杆菌对增强禽波氏杆菌OMP疫苗的免疫力有协同作用。

The synergistic effect of Taishan pine pollen polysaccharide and Lactobacillus on enhancing the immunity of subunit vaccine of Bordetella avium OMP

Abstract:In order to research the effect of immune enhancement induced by Taishan pine pollen polysaccharides and Lactobacillus. The modified method of water extraction and alcohol precipita- tion of Ma Rongchi Etc. was used to extract high-dose pine pollen polysaccharide. Three hundred one-day-chicks were randomly divided into six groups,group I , II , III ,and IV were immuned by subunit vaccine of Bordetella avium OMP with pine pollen polysaccharide at 100 g/L,group VI were immuned by subunit vaccine of Bordetella avium OMP without pine pollen polysaccharide. Group I and II were respectively orally given one milliliter of high and low dose of live Lactoba- cillus in the first two weeks,group m and IV were respectively orally given one milliliter high and low doses of heat-killed Lactobacillus in the first two weeks. Samples of blood and small intestine were taken every week after the first immunization. The level of systemic antibody was detected by indirect ELISA method,peripheral blood lymphocyte ratio was detected by automated hematol- ogy analyzer,peripheral blood lymphocyte transformation rate was detected by MTT method,in- testinal sIgA levels and serum IL-2 levels were detected by ELISA method. The results showed that each index in group I , II , HI and IV was significantly higher than that in group V and VI (P 〈0. 05) ;each index in group V was significantly higher than that in group VI (P〈0. 05) ; in the groups of live Lactobacillus was slightly higher than that in the groups of heat-killed Lactobacil- lus,in high dose groups were slightly higher than that in low dose groups. Therefore,pine pollen polysaccharide can significantly improve the immune effect of vaccine as the immune adjuvant,and oral administration of Lactobacillus can further enhance the animal immune response ability to the vaccine, the differences of each index between oral administration of live Lactobacillus and oral administration of heat-killed Lactobacillus were not significant. Pine pollen polysaccharide and Lactobacillus have a synergistic effect in enhancing the immunity of subunit vaccine of Bordetella avium OMP.
