小麦育种亲本SW3243中1BL/1RS易位的细胞学和分子标记检测及来源分析

摘    要:为了深入了解小麦育种亲本SW3243的遗传基础,利用多重PCR引物对SW3243及其他47个材料进行1BL/1RS易位系检测,同时对SW3243进行Giemsa-C带分析,并利用5对定位于黑麦染色体1RS上的特异PCR引物对1BL/1RS易位系进行了多样性分析。多重PCR分析结果表明,包括SW3243在内共23个材料含有1BL/1RS易位。细胞学研究结果证实,SW3243含有1对1BL/1RS易位染色体;筛选到定位于黑麦1RS上的3对SSR引物(Xmwg913、Xmwg2062a、SCM9)可用于区别不同的1BL/1RS易位染色体;用筛选到的3对特异PCR引物对SW3243及其他22个1BL/1RS品种(系)进行分析,结果表明SW3243所含1BL/1RS易位染色体与SW22514、西科麦2号、山前麦、川麦32、川麦35的1 BL/1 RS易位染色体不同。

Abstract:The plant area of SW3243 and its derived varieties were totally extended to 4.33 million hectare in Sichuan, and 7 varieties derived from SW3243 were released in Sichuan province and 3 were released in national level. Giemsa C banding displayed that dwarf, large spike and high yielding wheat breeding parent SW3243 had a pair of chromosomes 1RS/ 1BL. Moreover, 3 Specific PCR primers were selected to test SW3243 and other 22 varieties, Specific PCR testing indicated that SW3243 contained 1RS/ 1BL, but its 1RS/ 1BL was different from these 1RS/ 1BL contained by varieties SW22514, Xikemai 2, Predgornia, Chuanmai 32 and Chuanmai 35.
