| 摘 要： | 为制备抗鸡传染性喉气管炎病毒(ILTV)糖蛋白gD的单克隆抗体(MAb),本研究通过原核表达gD重组蛋白,纯化后免疫6周龄雌性BALB/c小鼠,经细胞融合筛选获得一株稳定分泌抗ILTV gD蛋白的杂交瘤细胞株,MAb亚型经鉴定为IgG1,轻链为κ链。Western blot结果显示,这株杂交瘤细胞分泌的MAb能够识别ILTV。ILTV gD蛋白的MAb的制备,为ILTV检测方法的建立奠定了基础。

| Abstract: | To prepare monoclonal antibody(MAb) against glycoprotein D of infectious laryngotracheitis virus(ILTV),the SP2/0 cells were fused with spleen cells of BALB/c mice immunized with the purified recombinant gD protein,a hybridoma cell line stably secreting antibody was obtained by scanning with indirect ELISA coating with purified ILTV as coating antigen.The MAb was identified to be IgG1 subclass with κ light chain.Western blot analysis showed that MAb reacted positively with ILTV.The MAb developed could serve as a tool for further study on ILTV and development of detection method for ILTV infection. |
