一株耐碱性芘降解菌的筛选及特性研究

摘    要:以长庆油田油井口附近的土壤为材料,芘为唯一碳源和能源的BH培养基中分离到一株高效芘降解菌株b2,经形态学观察和16SrDNA测定,将其鉴定为分枝杆菌属(Mycobacteriumsp.)。研究发现,在培养温度为37℃、转速为175r/min,培养起始pH 10,接种量为2.0%时菌体生长较快。最佳培养条件下,在芘质量浓度为250mg/L的BH培养基中,经菌株b2降解4d后质量浓度下降76%。说明培养条件对菌株的生长和芘降解速度有明显的影响。

Abstract:Using BH inorganic salt media which contain pyrene as sole carbon source and energy source,one PAHs-degradation bacteria is isolated from soil collected from Changqing oil field.The strain b2 is identified as Mycobacterium sp.through morphological observation and 16S rDNA.The strain b2 grows quickly under the conditions of 37 ℃,the initial pH 10 of media,shaker rotary speed 175 r/min,and inoculum amount 2.0%.Under the optimum growth conditions,76% pyrene can be degraded with the initial concentration 250 mg/L after 4 days.The growth of strain b2 is significantly affected by various culture conditions,so this bacteria has cheerful prospect for in-situ remediation of PAHs-contaminated soil.
