摘    要:利用PCR成功的从乙肝患者血清中扩增出乙肝表面抗原基因,并构建了含有此基因的植物表达载体pBHB。用根癌农杆菌(Agrobacterium tumefaciens)介导叶盘法将乙肝表面抗原基因转入百脉根(Lotus corniculatus),从转化植株中提取DNA做PCR检测、Southern杂交,阳性结果验证了目的基因已整合到百脉根的基因组中,转化效率为20%,证实转化的载体真实高效。

Abstract:To construct the high effect expression plasmid, the Hepatitis B surface antigen (HBsAg) gene was cloned from Hepatitis B virus DNA by using PCR method, and it was inserted into vector of pBin438 .The plant expression vector pBHB was obtained. Crowtoes(Lotus corniculatus)transformed by Agrobacterium-mediated method were analyzed for the integration of the transgene by PCR and Southern blot. The positive band indicated that the exterior gene had been integrated into crowtoe. There were 20% positive plants, which showed the expression vector pBHB was high effective.
