摘    要:研究了离体培养条件下不同浓度激素、活性炭、低温及其处理方式和种皮等因素对打破杜梨种胚休眠促进萌发的影响.结果表明,1/2MS培养基上附加GA3和6-BA可以有效地促进去皮杜梨种胚的萌发,以6-BA的促进萌发作用优于GA3,其中分别以7.5 mg/L 6-BA和15.0 mg/L GA3促进萌发和成苗效果较好,但根系发育以附加GA3的培养基为好,易发生侧根.低温和去种皮处理均有利于种胚的萌发,且表现接种后低温预培养方式优于低温处理后再接种培养.活性炭有利于侧根的发生,以0.1%水平促进萌发成苗作用较好;IAA具有很好的促进生根的作用,平均生根率可达89.54%,且以IAA为2 mg/L时侧根条数较多.

Abstract:The effect of different concentration GA3,6-BA and AC3 seed-coat, low temperature and treatment methods on breaking dormancy of embryo culture were studied with Pyrus betulaefolia Bunge.The result showed that the dormancy of embryo without seed-coat can be broken effectively by GA3 and 6-BA added in 1/2MS medium.The action of promoting germination of 6-BA is better than that of GA3,especially the germinating rate and plantlets forming rate were both high on 1/2MS with the addition of 7.5 mg/L 6-BA and 15.0 mg/L GA3.But GA3 added in 1/2MS is better for the root growth of plantlets,which occurred more lateral root.Lower temperature and removing seed coat can promote germination of embryo and the method of low temperature pre-culture after inoculation is prior to low temperature treatment before inoculation.AC can help lateral root sprout and the level of 0.1% was better for embryo germination and plantlet formation.IAA could promote rooting with average 89.54% rooting rate.
