摘    要:目的]探讨小麦种子萌发早期硫化氢(H2S)对淀粉酶活性的影响。方法]以NaHS作为H2S的供体,以扬麦158为供试材料,使用聚丙烯酰胺凝胶电泳法研究淀粉酶活性的变化。结果]使用H2S处理小麦种子可以有效提高总淀粉酶活性,当H2S浓度为0.4mmol/L,处理时间为9h时最佳。总淀粉酶中的β-淀粉酶活力增高而α-淀粉酶却不受影响。使用不同硫化物处理小麦种子9h,H2S处理过的小麦种子淀粉酶活性高于其他含硫化合物。结论]试验表明,H2S可以提高小麦种子体内β-淀粉酶活性,并呈剂量依赖性。

Abstract:Objective]The research aimed to investigate the role of endogenous hydrogen sulfide (H2S) to amylase activity in wheat seeds during early stage of germination. Method]NaHS was used as the H2S donor,seeds of wheat (Triticum aestivum L. ,Yangmai 158) were used for experimental material,and polyacrylamide gel electrophoresis was used to investigate the changes of amylase activity. Result]After treated with 0. 4 mmol/L H2S for 9 h,the amylase activity was improved effectively. β-amylase activity increased however α-amylase was not affected. After treated with different sulfide for 9 hours,the amylase activity of wheat seeds treated with H2S was much higher than other sulfur-containing. Conclusion]This experiment showed that H2S induced higher β-amylase activity in wheat seeds and it was dose-dependent.
