黄早四泛素连接酶Rbx1的分子克隆及原核表达

摘    要:通过RT-PCR方法和基因克隆技术对黄早四Rbx1基因进行分子克隆和序列分析,构建黄早四Rbx1的原核表达系统,成功表达出带有His标签的黄早四Rbx1融合蛋白,并进一步利用His抗体,用蛋白免疫印迹的方法对重组蛋白进行鉴定。结果表明,重组蛋白能够与His抗体发生免疫反应,验证了黄早四Rbx1在原核表达系统中成功表达。

Abstract:Molecular cloning and sequence analysis of Huangzaosi Rbx1 were carried out based on RT-PCR method and gene cloning technique. At the same time, the prokaryotic expression system of Huangzaosi Rbx1 was constructed and the recombinant protein with a His-tag was successfully expressed. Furthermore, western blot with His antibody was used to identify the recombinant protein. The results showed that the recombinant protein had immune reaction with His antibody, and verified that Rbx1 from Huangzaosi could be successfully expressed in prokaryotic expression system.
