东北地区野生百合遗传多样性的RAPD分析

摘    要:应用RAPD标记对中国东北地区6种3变种共34份野生百合试材的遗传多样性进行研究.筛选12个随机引物对其总DNA进行扩增,共扩增出101条带,其中83条带具多态性,多态性比率为82.1%,平均每个引物扩增出6.9条多态性谱带.基于 RAPD标记,利用 UPGMA构建了聚类树状图,34份野生百合资源的Nei&Li相异系数介于 0.013 7与0.601 3 之间.结果表明,中国东北地区野生百合具有丰富的遗传多样性,聚类树状图将34份试材分为2大类,第1类包括细叶百合和垂花百合,共5份试材,第2类包括卷丹、毛百合、有斑百合、大花百合、大花卷丹、朝鲜百合和东北百合,共29份试材;卷丹与有斑百合的亲缘关系较近,而与同为卷瓣组的朝鲜百合、细叶百合、大花卷丹等亲缘关系较远.细叶百合与垂花百合亲缘关系较近.

Abstract:The DNA polymorphism from 34 accessions of 6 species and 3 varieties in Lilium native to Northeastern China was studied using randomly amplified polymorphic DNA(RAPD) markers in the experiment.A total of 101 clear and reproducible bands were generated from twelve RAPD primers,of which 83 were polymorphic,with an average of 6.9 polymorphic bands per primer.The dendrogram based on RAPD data were constructed by using UPGMA analysis.The distance coefficient of the analyzed materials ranged from 0.013 7 to 0.601...
