湿地松合子胚发育与体胚诱导的研究

摘    要:以湿地松未成熟合子胚为材料,研究合子胚发育进程及影响体胚诱导的主要因素。结果表明,湿地松合子胚发育经历多胚阶段,即发育的第2、3阶段,此阶段的合子胚为体胚诱导最佳外植体,诱导率达30%;体胚诱导最佳培养基为LP+2,4-D 2.2 mg.L-1+6-BA 1.0 mg.L-1和DCR+NAA 2.0 mg.L-1+6-BA 0.63 mg.L-1+KT 0.61 mg.L-1,其体胚诱导率分别为24%和22.75%;球果短时间冷藏及培养基添加AgNO3、pH缓冲液均有利于提高体胚诱导率。

Abstract:With immature zygotic embryos (Zes) of slash pine as explants, the development course of Zes and main factors affecting initiation of somatic embryos (Ses) were studied. The results showed that Zes development went through polyembryony, while Zes was at development stage 2,3 that was the best explants. The Ses initiation rate of stage 2,3 Zes was 30%. The best initiation media were LP+2,4-D 2.2 mg·L~(-1)+6-BA 1.0 mg·L~(-1) and DCR+NAA 2.0 mg·L~(-1)+6-BA 0.63 mg·L~(-1)+KT 0.61 mg·L~(-1), in which the Ses initiation rates were 24% and 22.75%, respectively. Cold store of cones and media supplemented with AgNO_3 and pH buffer improved the Ses initiation rate.
