摘    要:【目的】探索种植密度对白肋烟烟叶游离氨基酸含量变化及品质的影响,确定白肋烟最佳种植密度,为优化其农艺措施提供参考。【方法】采用OPA、FMOC联合在线衍生反相高效液相色谱法对不同种植密度下白肋烟中游离氨基酸含量进行测定。【结果】种植密度对白肋烟氨基酸含量的影响表现为:天冬氨酸〉丝氨酸〉苯丙氨酸〉谷氨酸、甘氨酸、组氨酸、精氨酸+苏氨酸、丙氨酸、脯氨酸、酪氨酸、蛋氨酸、赖氨酸、亮氨酸〉缬氨酸〉异亮氨酸〉胱氨酸;总游离氨酸的含量随种植密度的增加呈先增加后减少的趋势;不同处理之间以行株距100cm×50cm、种植密度19995株/ha烟叶的香气质较纯净、香气量较足、杂气较少、刺激性较低、总得分最高,行株距110cm×45cm、种植密度20190株/ha和行株距120cm×40cm、种植密度20835株/ha次之,行株距120cm×50cm、种植密度16665株/ha得分最低。【结论】种植密度对白肋烟中的天冬氨酸、丝氨酸、苯丙氨酸含量的影响最大,而对胱氨酸、缬氨酸和异亮氨酸的影响较小。行株距110cm×45cm,种植密度20190株/ha时,总游离氨基酸含量最大,可获得质量好、品质优的白肋烟烟叶。

Abstract:[Objective]The present experiment was conducted to study the effect of different planting densities on the free amino acids content and quality of burley leaves and to determine the optimum planting density in order to optimize the cultivation technology for burley. [Method]The OPA/FMOC derivatized amino acids method using reversed phase high performance liquid chromatography was used to determine the free amino acids content in leaves of burley plants grown with different planting densities. [Result]The order of the effects of planting density on the contents of 17 free amino acids in burley leaves was as follows: AspSerPheGlu, Gly, His, Arg+Thr, Ala, Pro, Tyr, Met, Lys and LeuValIleCys-2. The total content of free amino acid increased initially, followed by a decrease with increasing planting density. The maximum content of total free amino acid was obtained in treatment 5 (row spacing 110 cm×45 cm, planting density 20190 plants/ha), followed by in treatment 6 (row spacing 120 cm×40 cm, planting density 20835 plants/ha). The minimum content of total free amino acid was obtained in treatment 9 (row spacing 100 cm×40 cm, planting density 25005 plants/ha). The burley leaves in treatment 4 (row spacing 100 cm×50 cm, planting density 19995 plants/ha) were found to have the best total score with nice aroma quality, high aroma quantity, lesser miscellaneous gas and weaker irritability. The lowest total score was found in treatment 1 (row spacing 120 cm×50 cm, planting density 16665 plants/ha). [Conclusion]The planting density highly affected the contents of Asp, Ser and Phe in leaves, while the contents of Val, Ile and Cys-2 were least influenced by planting density. The quality of burley leaves could be obtained by planting it in 110 cm×45 cm row spacing with 20190 plants/ha planting density.
