蚯蚓细胞色素P450基因的克隆及序列分析

摘    要:蚯蚓是一种土壤环境的指示生物,参与土壤中有毒化学物质的降解等过程。细胞色素P450酶是动物体内参与外源化合物代谢的主要酶系。本实验通过已有的其他物种的细胞色素P450酶保守序列设计简并引物,用RT-PCR的方法成功扩增出了赤子爱胜蚓体内的一条P450序列(670bp左右,Genbank登录号:KF823788),并对其保守区域、氨基酸构成及三级结构进行了分析。这是首次从蚯蚓体内扩增出P450基因,为下一步从基因层次研究蚯蚓机体对外源化合物的解毒功能及其机理提供了参考。

Abstract:Earthworm is a kind of bio-indicators for soil environment.They participate in the degradation of toxic chemicals in the soil.Cytochrome P450 (CYP450) is an important super enzymatic family in organisms with the detoxification function of xenobiotics.In this study,the primers were designed based on the conserved sequence of reported CYP450 gene from other animal species.One pieces of CYP450 gene was cloned by RT-PCR and then sequenced.It was about 670 bp and the accession number in Genbank was KF823788.The conserved sequence,amino acid composition and its tertiary structure were then analyzed.This is the first report about CYP450 gene clone from earthworm.This study is of important sense for the further study about detoxification of xenobiotics and its mechanism in earthworm body in the gene level.
