蛋白酶对曲霉原生质体稳定性再生及融合的影响

摘    要:4种蛋白酶对黑曲霉,米曲霉原生质体稳定性,再生有主融合的研究结果表明,蛋白酶用于原生质体灭活前或后保温均显著降低原生体数量,同时提高原生质体再生率及灭活性原生质体融合指数。蛋白酶安全,无毒可以替代常规促融合剂聚乙二醇。

Abstract:The effects of various proteinases on the storage and regeneration of normal protoplast and fusion of inactivated protoplasts between Aspergillus niger V. Tiegh and Aspergillus oryzae Ahlb were studied. The results showed that both proteinase incubation before and after inactivation reduced protolpast number and increased regeneration frequency of normal protoplast and fusion index of inactivated protoplasts. These effects of proteinase varied with treatment patterns such as incubation duration, proteinase concentration and combination of incubation with inactivation. It was observed that proteinase could replace polyethyleneglycol as a good alternative approach of common fusogen with a higher fusion index for inactivated protoplasts.
