大蒜抗草甘磷愈伤组织变异系的选择研究

摘    要:选取改良蒜、金堂早蒜、苍山蒜和欧引 0 1等 4个不同生态型的大蒜品种 ,采用直接接种和逐级培养法在质量分数 0 .0 2 % ,0 .0 4 % ,0 .0 6 % ,0 .0 8% ,0 .1 0 % ,0 .2 0 %的草甘磷培养基上 ,选择抗草甘磷愈伤组织细胞系。结果表明 ,草甘磷对大蒜外植体的出愈率有极强的抑制作用 ,在试验中均未诱导出愈伤组织。用愈伤组织直接在不同质量分数的草甘磷培养基上选择 ,4个品种在低质量分数 (<0 .0 4 % )胁迫下都有一定的抗性 ;经逐级抗草甘磷选择后的愈伤组织 ,其抗性能力有明显提高 ,在含有草甘磷质量分数 0 .0 6 %培养基上 ,4个品种愈伤组织的成活率为 1 0 .3%~ 1 8.2 % ,且发育良好 ;在质量分数 0 .0 8%和 0 .1 0 %的草甘磷培养基上也有部分愈伤组织成活。抗性稳定性检验证明 ,此抗草甘磷能力在一定时间内能够保持下去

Abstract:Four different ecotypic garlic cultivars:Gailiang,Jintang early,Cangshan and Europe 01 were chosen for this experiment.Their store leaf with part of stem plate and callus established from it was used to select glyphosate resistant on solid MS medium containing gradually increased concentrations of Glyphosate from 0.02%,0.04%,0.06%,0.08% to 0.10%.The results show that Glyphosate inhibited callus growth completely in all tested concentration levels;direct selection on medium containing different concentration of Glyphosate showed that callus of four cultivars all had Glyphosate resistant at lower concentration (<0.04%) of Glyphosate;Glyphosate resistant of callus enhanced obviously when selected by step to step increasing concentration of Glyphosate from MS medium.The survival rate of callus of four garlic cultivars on medium containing 0.06% glyphosate was in the range of 10.3%~18.2%;Mensuration of growth rate indicates that callus development was good.Some callus were survivor on medium containing 0.08% and 0.10% Glyphosate.Stability test of glyphosate resistant showed the induced glyphosate resistant could keep for a long time.
