不同浓度紫杉醇预处理对绵羊卵母细胞玻璃化冷冻保存效果的影响

摘    要:细胞骨架系统的稳定对于提高卵母细胞或胚胎冷冻后的存活率及其发育能力具有重要作用。紫杉醇(Taxol)作为细胞骨架稳定剂,可增强α、β-微管蛋白二聚体的紧密联系,增大微管交联,促进微管聚合和稳定。本实验以绵羊体外成熟卵母细胞为材料,旨在探讨紫杉醇预处理对玻璃化冷冻绵羊卵母细胞存活率、形态正常率及体外受精后胚胎发育率的影响。采用0、0.5、1.0、1.5μmol/L的紫杉醇预处理绵羊卵母细胞并进行玻璃化冷冻保存,未经处理的新鲜卵母细胞为对照组。结果表明:采用浓度为0.5μmol/L紫杉醇预处理绵羊卵母细胞冷冻保存效果最佳,其解冻后的存活率(82.38%)与其他3个试验组相比有显著差异性(P0.05);体外受精卵裂率(51.17%)和桑囊胚率(20.30%)也显著高于其他3个试验组(P0.05),但低于对照组(69.42%,34.77%)(P0.05)。由此可见,以0.5μmol/L浓度的紫杉醇预处理组绵羊体外成熟卵母细胞冷冻后能获得较好的效果。

Abstract:Stabilizing the cytoskeleton system during vitrification is beneficial for improving the post-thaw survival and subsequent development of vitrified oocytes or embryos.As a cytoskeleton stabilizer,taxol increases the rate of polymerization by reducing the critical concentration of tubulin needed for polymerization,and it stabilizes microtubules by causing tighter links between alpha and beta tubulin dimers and by enhancing microtubular cross-linking.Therefore,the in vitro matured ovine oocytes was used as the material in this experiment,to determine the effect of vitrification on survival rate,normal morphylogy rate,embryo development rates after in vitro fertilization by pretreating in vitro mature ovine oocytes with taxol.After pretreating with the concentration of taxol at 0,0.5,1.0,1.5 μmol/L respectively,the oocytes were vitrified.Untreated oocytes were used as control.The results showed that the survival rate(82.38%)in 0.5 μmol/L taxol group was significantly higher(P < 0.05) than those in other three groups;there was significant diferences between 0.5 μmol/L taxol group and control group in cleavage rate(51.37%vs.69.42%) and morula-blastocyst rate(20.30%vs.34.77%) of in vitro fertilization of frozen-thaw oocytes(P < 0.05),but was significantly higher(P < 0.05) than those in other three groups.In conclusion,0.5 μmol/L taxol pretreatment has positive effects on vitrified ovine oocytes matured in vitro.
