| 摘 要： | 高G C含量模板进行PCR扩增时,往往通过添加增强剂如甘油、DMSO、甲酰胺等来提高扩增效率,但是目前研究单个增强剂对PCR影响的较多,而研究增强剂组合的不多,而且关于增强剂对聚合酶保真度的影响的研究也不够充分。本研究通过正交试验对不同增强剂组合的效应进行了探讨,建立了3个对高G C含量模板进行PCR扩增的高效缓冲液体系,并且对其中2个进行研究发现它们可以提高Taq聚合酶扩增的保真度,因而具有较广的应用前景。

| Abstract: | Reagents such as glycerol,DMSO,formamide are usually used to improve the efficiency of PCR for the template with high G C PCR.Much of the work is focused on one or two reagents ' effect on PCR,rather than the combination of more reagents or the influence of reagents on the fidelity of DNA polymerase.Four reagents were recombined together using orthogonal experiment and three combinations were found efficient and two of them can also increase the fidelity of Taq polymerase. |
