桑树毛状根再生条件研究

摘    要:为建立桑树毛状根的再生体系,以桑树毛状根作为外植体,研究不同质量浓度6-BA和NAA配比对桑树毛状根愈伤组织诱导和不定芽分化的影响,并对再生植株进行PCR检测。结果表明,桑树毛状根的最佳愈伤组织诱导培养基为MS+0.2mg/L 6-BA+2.0mg/L NAA,最佳不定芽分化培养基为MS+1.0mg/L 6-BA+0.1mg/L NAA,3周愈伤组织不定芽分化率为16.7%。应用该不定芽获得再生植株的根系生长旺盛,叶片提取的基因组中含有rolB生根基因。

Abstract:To establish an asexual propagation system of mulberry hairy root,the effect of different concentration of plant growth regulators 6-BA and NAA on the mulberry hairy root growth in MS medium was studied.Also,PCR examination was carried out.The results showed that the best medium for callus induction was MS+0.2 mg/L 6-BA+2.0 mg/L NAA,and the best medium for adventitious buds induction was MS+1.0 mg/L 6-BA+0.1 mg/L NAA,with the ratio of adventitious buds induction of 16.7% within 3 weeks.The roots of plantlets from the adventitious buds were exuberant.PCR examination showed that rolB gene was successfully amplified from the leaf genome.
