蟾酥注射液对小鼠淋巴细胞影响的研究

摘    要:为了研究蟾酥注射液对机体淋巴细胞免疫功能的影响,选择不同浓度的蟾酥注射液作为试验组,分别与小鼠的脾淋巴细胞共同孵育48 h,检测细胞增殖水平的影响;用ELSIA法检测蟾酥注射液对几种重要的细胞因子(IFN-γ、IL-2和IL-12)分泌的影响;应用流式细胞技术检测蟾酥注射液对小鼠脾中T淋巴细胞CD4/CD8表达和细胞周期的影响。结果显示,蟾酥注射液在一定剂量范围内能够显著增强小鼠脾淋巴细胞的增殖能力;能够显著增强IFN-γ、IL-2和IL-12分泌;能够显著提高小鼠脾中CD4+ CD8-和CD4- CD8+表型的T淋巴细胞亚群百分率;能够显著提高小鼠脾中淋巴细胞进入S期的百分率。表明蟾酥注射液在体外能够显著提高淋巴细胞的增殖及功能,说明蟾酥注射能显著地增强机体特异性免疫功能。

Abstract:To study the effect of bufonis parenteral on lymphocytes, different concentration of bufonis parenteral solutions were selected as the experimental group. The BALB/c mice spleen lymphocyte incubated for 48 hours with different concentration of bufonis parenteral solution to test the level of cell proliferation. By ELISA method, the effects of bufonis parenteral on the secretion of several important cytokines (IFN-% IL-2 and IL-12) were evaluated. With the use of a method of flow cytometry tested the effect of bufonis parenteral solution on cell expression and cell cycle of CD4/CD8 of Tcell in mice spleen. The results indicated that bufonis parenteral solution in a certain dose range could significantly enhanced the proliferation of lymphocyte and the IFN-γ, IL-γ and IL-12 secretion; increase Tcell subset percentage of CD4^+ CD8^+ and CD4^+CD8^+ phenotypes in mice spleen and the percentage of lymphocyte entering into the phase. The results indicated that the bufonis parenteral solution could significantly increased the lymphocyte proliferation and function in vitro, concluded that bufonis parenteral solution has significantly enhanced specific immunological function.
