生长调节剂和外植体对油菜离体培养愈伤诱导的影响

摘    要:以双低优质油菜品种中双 7号的无菌苗为材料 ,探索了以子叶、下胚轴和幼根为外植体 ,及不同生长调节剂组合对愈伤诱导的影响作用。结果表明 ,生长调节剂是愈伤诱导成功的关键因子 ,2 ,4-D是诱导愈伤所必需的 ,以MS培养基含 1mg·L- 1 2 ,4-D与 2mg·L- 1 6 -BA配比为佳 ;同时子叶优于下胚轴和幼根作为外植体进行愈伤诱导 ,可获得出愈率高、增殖快、质量好的愈伤组织

Abstract:The effects of different growth regulator combinations and explants (cotyledon, hypocotyl and young root) upon callus induction of rape were studied, based on aseptically seedlings of "Zhongshuang No. 7", a double low elite variety The results indicated that growth regulator was crucial to induce callus of rape, in especial 2,4-D, the optimal induction medium which could induce callus was MS medium supplemented with 1 mg/L 2,4-D and 2 mg/L 6-BA, and as explants, cotyledon was fit for callus induction than hypocotyl and young root, to induce callus with high induction rate, quick growth and good quality
