Abstract:Spherica embryo, torpedo-shaped embryo and cotyledon embryo in loquat were used as explants of embryo culture.The experimental results showed that the highest ratio of callus and the optimum quality were obtained from cultured torpedo-shaped embryo. D-sorbitol as carbon source was beneficial for morphygenesis of cultured embryo, while sucrose as carbon source was efficient for the induction and multiplication of callus deried from embryo, and the callus was the optimum resource material for the isolation of protoplast. The best combination of enzymes selected for protoplast isolation was 2% pectinose with 1% cellualase, and with 12% mannitol. The yield of protoplast of 1×107/ g callus was obtained by using this conbination, and survival rate was above 95%. The first motisis was observed 3 days after that protoplasts were cultured in MS liquid medium added with 12% mannitol. And then motisising cell formed colonies, the latter further form littie callus in agrose medium. It was very difficulty for the callus derived from protoplast to dedifferentiate. After 8 months,the callus was transffered into the medium supplemented with 3% or 5% sorbitol, then shoot differentiation was observed first. Now several plantlets derived from protoplast were obtained.
