Abstract:The purpose of this study was to analyze proteomics in serum samples of cattle with different tuberculosis status.Tuberculosis positive cattle(TB,n=40)and healthy cattle(NC,n=20)were selected by the skin test(ST)and IFN-γrelease assay(IGRA).According to PCR testing for nasal swabs,40 TB cattle were then divided into PCR positive group(TBP,n=20)and PCR negative group(TBN,n=20).Serum samples were collected from all of these cattle and differential proteins were screened by isobaric tag for relative and absolute quantitation(iTRAQ)/mass spectrometry(MS).Besides,6 differential proteins were selected for parallel reaction monitoring(PRM)target verifi cation.The results showed that a total of 223 differentially expressed proteins were screened between TB and NC groups(FDR≥1.67 or≤0.67 and P≤0.05),among which 112 proteins were up-regulated and 111 proteins were down-regulated.A total of 74 differentially expressed proteins were selected between TBP and TBN groups,among which 46 proteins were up-regulated and 28 proteins were down-regulated.Parallel reaction monitoring was successfully quantitated to 6 validated proteins and the trend was consistent with iTRAQ.Bioinformatics analysis also indicated that the complement system and coagulation system signifi cantly changed after Mycobacterium bovis infection.The results of this study demonstrated that the great difference in serum protein compositions and physiological states existed among TB cattle with different infection statuses,which provided theoretical support for the distinction of different infection states of bovine tuberculosis.Therefore,the present study provided useful information for the prevention and control of bovine tuberculosis.
