诺氟沙星在鲫体内的药动学及残留研究

摘    要:研究了单剂量肌肉注射和多剂量混饲口灌给药方式下,诺氟沙星在鲫(Carassius auratus)体内的药物动力学和残留情况。结果显示:在(25.4±0.3)℃水温条件下,以每千克鱼体重10 mg的剂量单次给鲫肌肉注射诺氟沙星后,药物几乎在瞬间吸收,0.0333 h时血液中达到6.6708μg/mL,其血药浓度-时间数据用一级吸收二室模型描述较为合适,主要的药动学参数为:t1/2α、t1/2β、AUC和C l(s)分别为:0.4231 h、9.1613 h、17.8619μg.h/mL和0.5727 L/(kg.h)。在(23±1)℃水温条件下,以每千克鱼体重10 mg的剂量多次连续5 d混饲后,鲫停药后第8天肌肉、血清和肝脏中未检测到药物,此时肾脏中药物浓度已降到(0.0463±0.0134)μg/g,低于0.05μg/g。建议在(23±1)℃水温条件下,按每千克体重10 mg的剂量连续5 d混饲给鲫诺氟沙星,休药期至少为最后一次给药后的8 d。

Abstract:The present study investigated pharmacokinetics and residues of norfloxacin(NFX)in Carassius auratus.The drug concentrations in serum,muscle,liver and kidney were determined by Re-high performance liquid chromatography(Re-HPLC).The results showed that after a single-dosing intramuscular injection of 10 mg NFX/kg body weight in C.auratus at the water temperature of(25.4±0.3)℃,the drug concentration in the serum amounted to the largest(6.6708 μg/mL)immediately at the first sampling point at 0.033 h,the drug concentration-data in the serum can be described as a two-compartmental open model,the main pharmacokinetics parameters are t1/2α,t1/2β,AUC and Cl(s),which are 0.4231 h,9.1613 h,17.8619 μg·h/mL and 0.5727 L/(kg·h)respectively.And no drugs were detected in muscle,serum and liver in C.auratus,the concentration in kidney is(0.0463±0.0134)μg/g and lower than 0.05 μg/g on Day 8.Therefore,we suggest a withdrawal period should last at least for 8 days in C.auratus at the water temperature of(23±1)℃.
