鱼腥草根总黄酮的超声波辅助提取与体外抗氧化性研究

摘    要:通过单因素和正交试验对鱼腥草(Houttuynia Cordata)根总黄酮的超声波辅助提取工艺进行优化,采用ABTS·~+、·OH和H_2O_2清除试验,结合亚油酸乳化体系和猪油,研究了鱼腥草根总黄酮的体外抗氧化活性。结果表明,鱼腥草根总黄酮提取工艺为在超声功率200 W下,按固液比1∶60(g/m L),加入60%乙醇,55℃下提取35 min,总黄酮提取率达2.00%;鱼腥草根总黄酮的ABTS·+清除率随着浓度的升高而逐渐趋于稳定,·OH、H_2O_2清除率与浓度具有线性正相关性,均高于同浓度的VC溶液;鱼腥草根总黄酮可有效抑制亚油酸的过氧化反应,提高猪油的贮藏稳定性。综上所述,鱼腥草根总黄酮具有很好的体外抗氧化活性和脂质过氧化抑制效应。

Ultrasonic Assisted Extraction of Flavonoids from Houttuynia Cordata Roots and Its Antioxidation in Vitro

Abstract:The single factor and orthogonal design experiments were used to optimize the ultrasonic assisted extraction of flavonoids from Houttuynia Cordata roots and its antioxidant acitivities were evaluated in vitro by scavenging capabilities of ABTS·+,·OH and hydrogen peroxide and the lipid peroxidation inhibition in both linoleic acid-in-water emulsion and lard models.Results showed the highest extraction yield of flavonoids from Houttuynia Cordata roots could obtain to be 2.00%,using ethanol concentration of 60%(v/v) as feed-to-liquid ratio of 1:60 (g/mL) extracted for 35 min at 55℃ assisted with ultra-sound of 200 W. The ABTS·+ scavenging rate of flavonoids was improved to a stable value with the increasing concentration. The ·OH and hydrogen peroxide scavenging rates of flavonoids were significantly positive correlation with concentration,and both higher than Vc solution with the same concentration. Lipid peroxidation in linoleic acid was effectively inhibited by flavonoids,as well as the storage stability of lard was improved. It was indicated that the flavonoids from Houttuynia Cordata roots possessed good vitro antioxidation and lipid peroxidation inhibition.
