不同成熟度烤烟叶细胞化学研究

摘    要:采用酶解方法分离K326不同成熟度烟叶中部叶(第10叶)栅栏细胞和海绵细胞,运用气相色谱分析鉴定细胞中非挥发性有机酸,分光光度计法分析质体色素、糖类及烟碱.发现多元有机酸在海绵细胞中含量较高,超过栅栏细胞的2倍以上,高级脂肪酸、质体色素、糖类均是栅栏细胞中含量高,总量分别超过海绵细胞的2,1.2,1.2倍.而海绵细胞烟碱含量高于栅栏细胞的2倍以上;在烤烟成熟过程中,有机酸、叶绿素和类胡萝卜素的含量逐渐降低,过熟海绵细胞有机酸、叶绿素和类胡萝卜素含量分别仅为未熟的50%,16%,50%.过熟栅栏细胞有机酸、叶绿素和类胡萝卜素含量仅为未熟的50%,20%,55%.糖类在欠熟细胞内含量最高,过熟细胞内最低,欠熟海绵细胞为过熟的1.6倍,欠熟栅栏细胞为过熟的2.4倍.烟碱随着成熟度增加而逐渐积累,适熟细胞内含量最高,未熟细胞内最低.适熟海绵细胞为未熟的2.8倍,适熟栅栏细胞为未熟的5.4倍.

Abstract:The spongy and palisade cells of different maturity cutter leaf(10th) of Flue-cured Tobacco K326 were divided by pectinase,then no-volatility organic acid was detected by gas chromatography, and plastid pigment or glucide or nicotine was analyzed by spectrophotometer.It showed that the content of multiple organic acid and nicotine of spongy cells was higher than that of palisade cells while the content was 2 times and other components were on the contrary.The content of organic acid,pigment,sugar was 2,1.2,1.2 times respectively.With the maturing,organic acid,chlorophyllous and carotin depressed gradually.The content of organic acid, chlorophyll and carotenoid in overripe spongy cells was 50%,16%,50% of unripe spongy cells.The content of organic acid, chlorophyll and carotenoid in overripe palisade cells was 50%,20%,55% of unripe palisade cells.With the most content of glucide in maturity cells , the content of glucide in unripe spongy cells was 1.6 times as much as overripe cells,and the content of glucide in unripe palisade cells was 2.4 times as much as overripe cells.The content of nicotine accumulated with the maturing.The content of nicotine in ripe spongy cells was 2.8 times as much as unripe cells,and the content of nicotine in ripe palisade cells was 5.4 times as much as unripe cells.
