摘    要:目的 以核糖体转录间隔区(rDNA ITS)序列为分子标记,对一株市售人参属植物归类到种提供分子证据.方法 改良CTAB法提取植物总DNA,利用通用引物ITS5/ITS4扩增rDNA ITS序列,经克隆、测序后,运用Clustal X,BioEdit和PAUP等软件进行序列分析并构建系统发育树.结果 测序得到该植物的rDNA ITS区序列长度为722bp,序列分析结果显示该植物与Genbank中已有的人参(PanaxGingsen,FJ593178)rDNA ITS区序列之间相似性达100%,在系统发育树中并排聚类成一枝.结论 基于rDNA ITS区序列的测序分析和系统发育树构建的分子生物学方法,能够对供试品进行准确的分子鉴定.

Abstract:Objective To establish the molecular method to identify the Panax Ginsen from Changbaishan of Jilin provience.Methods Total DNA was extracted with modified CTAB method and thereby rDNA ITS regions were amplified with general primer ITS5-ITS4,sequencing and phylogenetic analyses using Clustal X、BioEdit and PAUP.Results The rDNA ITS entire sequence of the sample was 722 bp,sequence analysis shows that the rDNA ITS sequences similarity with Panax Gingsen(FJ593178) is 100%.The sequence is closely related to the genus of Panax Gingsen in GenBank.Conclusion Based on molecular biology methods of rDNA ITS region analysis,molecular identification is available in accurate classification of market sold Panax Gingsen.
