摘    要:目的 探讨红花茎尖微嫁接技术,为解决转基因红花植株生根困难问题提供一个新的途径.方法 光照条件下培养红花实生苗用作砧木,以红花组培苗茎尖为接穗,采用劈接法和顶接法进行茎尖微嫁接试验,并对影响嫁接成活率的因素进行研究.结果 成功获得了红花茎尖微嫁接苗,并优化了嫁接的条件:选取苗龄为14d的红花实生苗为砧木,嫁接成活率较高;采用劈接法进行茎尖微嫁接,效果较好,嫁接成活率可达56.7%,而顶接法成活率仅为16.7%.结论 本研究建立了红花茎尖微嫁接的方法,获得了嫁接成活苗.

Abstract:To study the technique for shoot-tip grafting of safflower in order to support a new method for regeneration of transgenic safflower root.Using light grown safflower seedling as rootstock and shoot-tip of safflower as scion,the shoot-tip grafting was executed by grafting a scion of safflower tissue culture seedling using cleft grafting and top grafting method.And the factors affecting the survival rate of shoot-tip grafting were studied.The micro-grafted plantlets of safflower were achieved.It was suggested that the more survival rate of grafting was obtained by using 14 day old light grown seedlings as rootstocks.The eleft grafting method was optimal for grafting,with survival rate of 56.7%,but the survival rate of top grafting method is 16.7%.In this study,a procedure of shoot-tip grafting in vitro has been established in safflower and micro-grafted survival plantlet s have been obtained.
