马铃薯试管苗保存毒源PVA技术的研究

摘    要:报道了含PVA毒源的马铃薯试管苗保存技术及效果。在无菌条件下切取毒源马铃薯茎尖于盛MS培养基的试管中培养,当毒源马铃薯长至12 cm时单节切段进一步扩繁培养。利用免疫电镜法、电镜负染法筛选出3管纯毒源材料。进一步试验表明:免疫电镜法鉴定证明毒源马铃薯试管苗接种3周后病毒含量明显提高;毒源马铃薯试管苗移至花盆中栽培表现出典型的PVA症;用毒源马铃薯的汁液接种指示植物表现典型PVA状,说明毒源马铃薯试管苗的PVA具有侵染力。用马铃薯试管苗保存PVA毒源,克服了传统保存中的不足,提高了保存效果。

Abstract:The paper introduces the technique and preservation method of test-tube potato plantlet infected by Potato Leaf-roll Virus A(PVA) and its effects.The test was done like this: cut the stem tips of potato plantlet with PVA in the state of asepsis and cultivate them in the tube with MS,further culture them by cutting them into single sections when the stem tips grow to 12 cm.Select 3 tubes of potato plantlet infected by PVA through immunity electronic microscope capturing and electronic microscope negative staining.Further test identification by electronic microscope shows a high PVA content 3 weeks after the plantlets' inoculation;test-tube potato plantlet infected by PVA planted in the flower pot shows typical symptom of PVA The indicator plants globeamaranth,The typical symptom of PVA,after being inoculated with the potato infected by PVA.It shows that PVA in test-tube plantlet has infectious ability.To preserve PVA in potato test-tube overcomes the shortcomings of traditional preservation methods,improves the preservation effects.
