山薯组培苗瓶内与瓶外生根技术研究

摘    要:以山薯组培苗茎段为外植体,进行组培苗瓶内与瓶外生根技术研究。结果表明,山薯组培苗茎段瓶内生根较优培养基配方为1/2MS液体培养基附加0.5 mg/L NAA、0.1 mg/L 6-BA、30.0 g/L蔗糖和1.5 g/L活性炭;在不同月份移栽对生根苗成活率影响较大,其中以4、10月的移栽生根苗成活率显著高于7月;采用瓶外生根方法,山薯组培苗茎段可不经任何生长素处理,经过21 d的培养,即可获得近90%的生根率;瓶外生根苗培养6周后可在基部形成1~3个微型薯,6个月后可形成1.5~4.0 g大小的微型薯。

Abstract:The in vitro and ex vitro rooting of D.fordii variety Guihuai No.2 has been carried out using stem of plantlets as explants.The results showed that 1/2MS basic medium supplemented with 0.5 mg/L NAA,0.1 mg/L 6-BA,30.0 g/L sucrose and 1.5 g/L AC in liquid culture was the preferred medium for in vitro rooting.The survival of rooting plantlet was affected by transplanting in different seasons,and found to be higher in April compared to July.Using ex vitro rooting technique,the stem of plantlet were found to hav...
