摘    要:本研究以影响番茄遗传转化的4个因子基因型、外植体类型、菌液浓度和是否添加AS进行了试验,结果表明不同基因型间、不同菌液浓度间、是否添加AS对番茄抗性愈伤率的影响都存在显著差异,以东农704,菌液浓度为0.6~0.8,添加AS有利于番茄的遗传转化。以LycE为目的基因,应用根癌农杆菌介导法对番茄进行了遗传转化,获得了完整的抗性植株,有3株经PCR和PCR-Southern分析鉴定,初步证明外源基因LycE已整合到番茄的基因组中。

Abstract:Four factors including genotype, the concentrations of bacterial suspension, explants and adding AS or not which effect the genetic transformation of tomato had been studied in this paper. The results showed that the effect of different genotype, different concentrations of bacterial suspension and adding AS or not on frequency of resistant callus inducing in tomato had significant differences. Using Dongnong 704 species as acceptor, the suitable OD600 of bacterial suspension was 0.6-0.8, and the addition of AS were beneficial to the genetic transformation of tomato. Then Lyre gene was transformed into tomato via agrobacterium tumefaciens. Resistant regeneration plants of tomato were obtained in selective condition. PCR and PCR - Southern analysis of 3 resistant plants originally proved that Lyre has been transferred into genome of tomato.
