摘    要:探讨Agilent 2100 Bioanalyzer芯片分析系统在检测食源致病性沙门氏菌的应用.根据致病性沙门氏菌的致病基因invA设计特异性引物扩增出致病性沙门氏菌特异性序列,然后分别用常规的琼脂糖电泳技术和Bioanalyzer对PCR产物进行检测,并比较两种检测方法的准确度、重复性和灵敏度.Bioanalyzer的准确度和重复性比琼脂糖凝胶电泳高,Bioanalyzer的准确度在95%以上,琼脂糖凝胶电泳仅为85%;Bioanalyzer的检测灵敏度也比琼脂糖凝胶电泳高.Bioanalyzer芯片分析系统结合PCR方法可对食源性致病菌进行特异、准确、稳定、灵敏的检测和鉴定,具有重要的推广价值.

Abstract:To explore the feasibility of using Agilent 2100 Bioanalyzer in the detection of foodborne pathogens Salmonella.The inv A gene of Salmonella spp.was amplified by PCR technique using specific primers.A gilent 2100 Bioanalyzer and conventional agarose gel electrophoresis were employed respectively to examine the PCR products,and accuracy,reproducibility and sensitivity between these two detection techniques were compared.Agilent 2100 Bioanalyzer showed more accurate in determining the length of the gene fragments than agarose gel electrophoresis,the accuracy of the former reached above 95% while the latter was only 85%.Bioanalyzer was also more sensitive than agarose electrophoresis.Agilent 2100 Bioanalyzer combined with PCR is specific,accurate and sensitive for Salmonella detection.
