大蒜胚性细胞悬浮系的建立

摘    要:以中牟 5号大蒜根尖为外植体 ,在 1 1种固体培养基上诱导愈伤组织 ,筛选出最佳培养基MS + 2 ,4-D 0 .5mg/L和MS + 2 ,4-D 2 .0mg/L +KT 0 .5mg/L。愈伤组织形成后在上述1 1种培养基上继代 3~ 4次 ,获得胚性愈伤组织后再转至液体培养基中 ,经强化培养 3个月后获得胚性细胞悬浮系。试验结果表明 :缩短换液时间 (每隔 3~ 4天换液 1次 ) ,可加快胚性细胞悬浮系的建立

Abstract:Root tips of the garlic were used as explants in inducement of garlic calli on the ll lcinds of solid media MS+2,4-D 2.0 mg/L+KT 0.5 mg/L (MS 2) and MS+2,4-D 0.5 mg/L (MS 9) was found to be the best media for callus growth.The embryogenesis calli were gained after the formed calli were put on 11 kinds of media for continued culture in 3~4 generations. The embryogenic cell suspension culture line was set up after the calli were transferred to the liquid medium MS-2 and MS-9 for 3 months of intensive clture.The results of the test show that the embryogenic cell suspension culture can be construsted quickly by shortening the period of changing media,e.g changing media every 3 or 4 days.
