摘    要:腺病毒载体具有高效传递和表达外源基因的能力,常被用作体内和体外基因转移或表达的工具.体内同源重组和体外直接连接是目前构建腺病毒重组体的两种主要方式.口蹄疫病毒基因重组腺病毒活载体疫苗多是以缺失E1和E3基因编码区的人5型腺病毒AdHu5为载体进行构建,有些载体还缺失了E4基因编码区.研究证实,已构建的口蹄疫重组腺病毒疫苗对猪、牛都有一定的免疫保护效果,在口蹄疫预防研究领域具有很好的应用前景.

Abstract:Adenoviral vector is a useful tool for achievement of highly efficient exogenous gene delivery and expression in vivo and in vitro.Recombinant adenovirus is usually constructed by in vivo homologous recombination or by directly in vitro ligation at present.Adenoviral vectors are also used for development of live vector vaccine of FMD,and in most vectors,E1and E3 gene coding region were deleted and sometimes E4 gene coding region were deleted from the AdHu5 genome.Currently,the recombinant adenovirus that expresses of FMDV empty capsid were proved to be very effective for delivery of protection against homologous virus challenge in swine and cattle.
