牛奶尿素氮测定方法研究

摘    要:本试验结合二乙酰-肟试剂盒法探讨牛奶尿素氮的测定方法。结果表明:对牛奶样品进行1500×g离心10min脱脂,再3500×g离心20min脱蛋白的前处理与不脱脂,3500×g离心20min脱蛋白的前处理相比,牛奶尿素氮测定值差异极显著(P<0.01),与不脱脂,5000×g离心20min脱蛋白的前处理相比,牛奶尿素氮测定值差异不显著(P>0.05)。牛奶样品不经过脱脂过程,5000×g离心10min和15min与离心20min牛奶尿素氮测定值差异不显著(P>0.05),但离心10min的牛奶尿素氮测定值不稳定。研究结果表明,牛奶样品可以不脱脂,经5000×g离心15min脱蛋白后测定牛奶尿素氮含量。

Abstract:The experiment was conducted to discuss the measurement of milk urea nitrogen ( MUN) with diacetyl-oxime kit. The result showed that centrifuging at 1,500×g for 10 mins to degrease and at 3,500×g for 20 mins to deprotein had a significant difference in not degreasing and centrifuging at 3,500×g for 20 mins to deprotein, and there was not significant difference in not degreasing and centrifuging at 5,000 ×g for 20 mins to deprotein. There was not significant difference between centrifuging at 5,000×g for 10 mins,15 mins and 20 mins, but the results of centrifuging for 10 mins were unstable. These results indicated that centrifuging at 5,000×g for 15 mins and not degreasing could measure content of MUN.
