沙冬青cDNA-AFLP体系的建立及引物筛选

摘    要:目的]建立适合于沙冬青cDNA—AFLP的反应体系。方法]以对照、3种逆境处理的沙冬青为材料,采用Trizol法和改良CTAB法提取总RNA。结果]改良的CTAB法可抽提到较高质量的总RNA,适于沙冬青总RNA的提取。采用EcoRI/MseI酶切体系,64对选择性扩增引物中有35对适合沙冬青cDNA-AFLP分析。结论]该研究为利用cDNA-AFLP标记对沙冬青抗逆基因进行深入研究奠定基础。

Abstract:Objective] The research aimed to establish the cDNA-AFLP system in Ammopiptanthus mongolicus.Method] RNA was extracted from the control and three stress-treatment materials.Trizol and CTAB method to extract RNA were compared.Result] Improved CTAB method could obtain a higher quality of the total RNA in Ammopiptanthus mongolicus.Using EcoR Ⅰ /Mse Ⅰ digestion system,35 selective amplification primer pairs were identified from 64 selective amplification primer pairs,which were suitable for cDNA-AFLP analysis.Conclusion] The research could provide important evidence for further studying the stress-response genes in Ammopiptanthus mongolicus.
