摘    要:本试验分别采用常规石蜡切片、脂肪包卵石蜡切片和冰冻切片3种方法对意大利蜜蜂卵进行切片HE染色和显微镜观察。结果发现:(1)常规石蜡切片法难以制作出蜜蜂卵切片。(2)冰冻切片法可以制作出清晰完好的蜜蜂卵组织切片,对样本中的蛋白质和核酸保存较好,但也有切片较厚,难以连续切片,保存时间短等固有缺陷。(3)脂肪包卵石蜡切片制作法能制作出完整而清晰的蜜蜂卵组织切片,且切片较薄,卵切片组织的清晰度和完整度要优于冷冻切片,该方法制作出的卵切片也利于切片的长期保存,但卵的整体外观形态存在一定程度缢缩。

Comparison of Three Methods for Honeybee Egg Histological Section
Abstract:In the present study,honeybee eggs were collected to make histological sections,three different methods were taken including normal paraffin production method,the method of paraffin production with fat en?folding,and the frozen section procedure.The sections were observed under microscope after finishing section procedure and dying by HE.The results showed that:1) it was hard to successfully gain the paraffin sections for honeybee egg because of its tiny size;2) it was suitable to produce honeybee egg sections using frozen section procedure,which was a common method to make sections for tiny tissue samples;however,there were some ob?vious shortages,such as short conservation time,likely damage to tissue structure;3) paraffin production with fat enfolding could greatly help make clear,thin and intact honeybee egg sections,which were better than fro?zen sections and suitable for long time conservation and further usage.However,there was some extent of con?striction along the shape of honeybee egg in the paraffin section.
