油菜MICMS恢复基因近等基因系的构建与近等性分析

摘    要:以宁R7为恢复基因源,利用连续回交法构建以T78、T84和ZS9三个优异种质为遗传背景供体的近等基因系候选株系。于成熟期考察并比较候选株系农艺性状,同时采用SSR标记对候选株系进行PCR扩增估算遗传相似系数并进行聚类分析。农艺性状分析(t测验)表明,不同候选株系与其供体的农艺性状具有不同程度的相似性,株高和分支点高度的差异程度较其他性状高。采用75对SSR引物对21个候选株系PCR扩增共得到157个目的片段,发现三个遗传背景候选株系的平均遗传相似系数分别为0.86、0.92和0.98。农艺性状与SSR标记综合分析后,认为wh13、wh59和wh118分别为三个种质的最优近等基因系。综上表明,采用农艺性状比较与分子标记相结合分析近等性是一种有效的近等基因系的检测方法。中选近等基因系可用于恢复基因的定位及克隆。

Abstract:This research aims to breed the restorer gene near-isogenic lines of the MI CMS system and explore the molecular mechanism of the restorer gene. With NingR7 as the restorer gene resource, the restorer gene candidate NILs which had the genetic background of three outstanding germplasms (T78, T84 and ZS9) were constructed by using the continuous-backcross method.
