苹果种质资源玻璃化法超低温保存技术

摘    要:研究了苹果茎的玻璃化法超低温保存技术。实现了低温锻炼、预培养及恢复培养基对成活率的影响,筛选出了合适的冰冻保护剂PVS3,构建了苹果茎玻璃化法超低温保存体系。同时对冻存成活苗的叶片再生能力和过氧化物酶(POD)同工酶进行了检测。

Abstract:The present essay studies the cryopreservation technique of apple shoot tips by vitrification.By experiments,the effects of cold hardening,preculture and recovery media on survival rate were tested.An optimal cryoprotectan(PVS 3)was selected and a method of the vitrification cryopreservation procedure of apple shoot tips was constructed.In addition,the leaf regeneration ability and peroxidase isozyme of the survival shoots were examined.
